Frequently asked questions for: Citric Acid Test

FAQ1: Can the Citric Acid Test be performed on frozen/thawed semen plasma samples?
Yes, it can. It is best to centrifuge the semen sample before freezing the semen plasma at -18°C or below.

FAQ2: Is it normal that precipitations are formed after adding reagent 2?
Yes, reagent 2 (isopropanol) will remove spermatozoa and particles by forming a precipitate. The extent of particle forming is dependent on sperm concentration and the number of particles/debris in the sample. Therefore, it is recommended to perform the test on seminal plasma (some precipitation is still present, but this is normal). It is very important to avoid aspiration of the precipitates when 25 μl supernatant is taken after centrifugation.
